Knights: Veteran back-rower Adam Elliott returns after missing Round 1 and takes over on the bench from Brodie Jones, who is listed as 18th man. No other changes for Adam O'Brien after a solid first-up win over Wests Tigers.
Dolphins: The loss of Kulikefu Finefeuiaki (concussion protocols) and Connelly Lemuelu (fractured eye socket) forces a restructure in the pack with Oryn Keeley promoted from 18th man to start in the back row and Mark Nicholls joining the bench in his return after missing last week's game to remain in Queensland with his family during the cyclone. Five-eighth Kodi Nikorima also returns to the starting side so Sean O'Sullivan drops to the reserves.

Panthers: Star winger Brian To'o (hamstring) lines up for his first game of the season so Paul Alamoti shifts to the centre and Luke Garner reverts to the bench. On the other wing Kiwi Casey McLean has recovered from a head knock suffered in Round 1 at Allegiant Stadium and will take his place. Garner's move to the bench pushes Luron Patea to the reserves. Also among the reserves is Liam Henry, who is nearing a return from a shoulder injury.
Roosters: With Victor Radley (concussion protocol) sidelined, last week's debutant Salesi Foketi has been promoted to the starting side at lock and former Eel Makahesi Makatoa has been added to the bench. Injury-plagued centre Billy Smith has completed successful hip and ankle rehabilitation and is listed among the reserves. Hugo Savala is again the 18th man. Star forward Spencer Leniu will return in Round 3 from suspension.

Bulldogs: Josh Curran has served his one-match ban incurred in Round 3 of the Pre-Season Challenge and comes in on the bench for Harry Hayes, who shifts to 18th man. Kurt Mann is again named to start at lock but may swap with Jaeman Salmon as he did last week an hour before kick-off.
Titans: Coach Des Hasler's first team for 2025 features big-name recruit Reagan Campbell-Gillard in the front row alongside Moe Fotuaika with skipper Tino Fa'asuamaleaui at lock. RCG has overcome a shoulder injury to take his place while big Tino tastes his first NRL action since rupturing his ACL in Round 3 last year. AJ Brimson and Jayden Campbell team up in the halves as they did in the final trial game against the Dolphins. David Fifita and Jaimin Jolliffe come off the bench along with Chris Randall and former Rabbitoh Brock Gray, who arrived at the club on a train and trial deal with 129 NSW Cup games to his name.
